ACETALDEHYDE (CH3CHO) is one of the most widely occurring aldehydes in nature and is commonly used in industry. Acetaldehyde, a metabolic byproduct of ethanol in the liver, is toxic to the human body and is rapidly converted to the less harmful acetic acid by the enzyme aldehyde dehydrogenase. People with a deficiency of aldehyde dehydrogenase accumulate acetaldehyde when consuming alcohol and this accumulation results in facial and body flushing often referred to as “Asian flush syndrome.” Build up of acetaldehyde has also been associated with the effects of hangovers from alcohol consumption. Although classified as a carcinogen, acetaldehyde is naturally found in many foods and beverages such as ripe fruit, coffee, and wine. BioAssay Systems colorimetric acetaldehyde assay kit is based on aldehyde dehydrogenase catalyzed oxidation of acetaldehyde, in which the formed NADH reduces a formazan reagent. The intensity of the product color, measured at 565 nm, is directly proportional to the acetaldehyde concentration in the sample.How specific is this assay for acetaldehyde?
The assay mechanism uses the enzyme aldehyde dehydrogenase to quantify acetaldehyde in the sample. The enzyme will also oxidize other aldehydes in the sample (i.e. propionaldehyde). Therefore, the assay will not distinguish between acetaldehyde and other aldehydes present in the sample. Consideration should be taken if high levels of other aldehydes are expected in the sample being tested.
How long are the diluted standards good for?
Acetaldehyde is a rather volatile chemical, so proper storage of the standard is vital. Only bring the 3 M Standard to room temperature for time needed to prepare standards. Return to -20°C within 30 minutes of thawing. Be sure to keep the Standard and standard dilutions on ice or refrigerated with the tube caps closed during assay procedure. The diluted standards for the colorimetric assay (4 mM, 2.4 mM, 1.2 mM, and 0 mM) and the fluorimetric assay (60 µM, 36 µM, 18 µM, and 0 µM) should be prepared fresh before each assay and used within 10 minutes of preparing.

The assay may be run at room temperature (25°C) or 37°C.
